Doctoral student (m/f/d) in the field of “Functional analysis of hierarchically organized p53 phosphorylation networks“

Support us from as of now in part-time (50 %) as a Doctoral student (m/f/d) in the field of “Functional analysis of hierarchically organized p53 phosphorylation networks“

In accordance with § 2 WissZeitVG and § 72 HessHG, the position is to be filled on a fixed-term basis at the Chair of Signal Transduction at the Institute of Biochemistry at the Faculty of Medicine with the opportunity for own academic qualification. The salary is in accordance with the collective labour agreement of the State of Hessen (E 13 TV-H).

As long as the maximum permissible duration of a fixed-term contract is not exceeded, you will be employed for an initial period of 3 years. An extension for up to another 2 years is possible under the above-mentioned condition.

Our research group focuses on mechanisms of signal transduction and inducible gene expression. In this context, we also study the transcription factor p53, one of the central switch points of cellular homeostasis. The p53 protein is activated by numerous stress signals, in this context we have discovered a novel DNA damage-inducible p53 phosphorylation site in the C-terminal oligomerization domain. This phosphorylation site acts as a master switch that allows the induction of further subordinate phosphorylations at different positions of the p53 protein. Accordingly, the absence of phosphorylation at the master site leads to a defect of all important p53 functions. In addition, relevant kinases for phosphorylation at this higher-order site will be identified. Further experiments will examine the effects of master site phosphorylation on the conformation of modified p53 and identify the molecular mechanisms that make this phosphorylation the master switch for further downstream phosphory­lations. ChIP-seq experiments will examine the effects of this phosphorylation on the chromatin association of p53. It will also be important to analyze the effects of oncogenic point mutations on p53 phosphorylation patterns. Together, these experiments will lead to a deeper understanding of p53 regulation and may potentially open further options to modulate disease-relevant p53 activities. This challenging project involves a wide range of different methods in the field of biochemistry and molecular biology.
